The rational hapten design and development of an indirect enzyme-linked immunosorbent assay (ELISA) for the detection of 4-tert-octylphenol (4-tert-OP), an endocrine-disrupting compound belonging to alkylphenols (APs), is described. The polyclonal antibody highly specific to 4-tert-OP was raised against a hapten with a linker attached to the ortho- position of the phenol ring. Using 5-(2-hydroxy-5-nonylphenyl)-5-oxo-pentanoic acid (NP1) as a competing hapten, an indirect competitive assay exhibiting an IC50 value of 100 mu g L-1 and the limit of detection (LOD) of 10 mu g L-1 was developed. The assay was highly specific to 4-tert-OP and only displayed a moderate cross-reactivity (29 %) with IGEPALA (R) CA-520. The immunodominance was suggested to be both the 4-tert-octyl chain and the phenol group. The assay was optimized for the analysis of 4-tert-OP in prawn and clam samples with the LOD of 20 mu g L-1 and the % recovery of 80-120 %.
