A method was developed for the determination of N,N-dimethylacetamide (DMAC) and N-methylacetamide (NMAC) in human urine by activate carbon solid phase extraction followed with gas chromatography. mass spectrometry. DMAC and NMAC were concentrated and cleaned up by activated carbon solid phase extraction column under the condition of pH 7. 0 and eluted with methanol. Targeted compounds were determined by gas chromatography-mass spectrometry (selected ion mode) with innowax capillary column. Under the optimized conditions, the calibration curves of DMAC and NMAC were linear in the ranges of 0. 1-40. 0 mg/L. Correlation coefficients were 0. 9997 and 0. 9995, respectively. The mean recoveries of DMAC and NMAC in spiked urine samples at the three concentrations (1. 0, 5. 0 and 25. 0 mg/L) were 94. 7%-102. 6% and 96. 6%-101. 2% with the relative standard deviation (RSD) less than 5. 5% (n = 6), respectively. The detection limits (S/N = 3) were 0. 01 mg/L and 0. 03 mg/L for DMAC and NMAC, respectively. The proposed method has high sensitivity and resolution. This may also provide an accurate and stable way of monitoring the amount of N,N-dimethylacetamide and N-methylacetamide in human urine.
