A spectral surface plasmon resonance (SPR) biosensor has been used to determine the hepatitis B surface antigen (HBsAg). The anti-HBsAg monoclonal antibody was immobilized on the surface of the sensor chip using the technology of self-assembly and a bifunctional cross-linker. The detection limit of HBsAg solution was 0.06 ng/mL. The results of responses to HBsAg control serum and to pure HBsAg solution showed that the selectivity of SPR biosensor was good. The results of experiments showed that the detection limit of the SPR biosensor was much lower than that of enzyme-linked immuosorbent assay (ELISA) method.
