Little attention has been paid to the detection and effects of anaerobes like Clostridium pelfringens (with high prevalence in swine industry) in boar sperm. The aim of this work is to detect the presence of C perfringens in boar semen doses destined to artificial insemination (AI) procedures to avoid the use of doses with low sperm quality, as well the spread of this bacterium. For this purpose, 9 mL of semen diluted (1:6) with a BTS-based extender containing kanamicine, obtained from commercial doses of healthy and sexually mature Pietrain boars were infected with different C perfringens strains at eight different concentrations. After DNA extraction, two conventional PCRs and a multiplex PCR were performed to detect all types of C. perfringens. The PCRs were useful to detect C pelfringens in boar semen up to a concentration of 101 bacteria/mL. In conclusion, C. perfringens can survive in boar semen samples and PCR techniques are valuable to detect this bacterium and avoid the use of doses with diminished sperm quality in boar semen samples. Moreover, the detection would avoid the spread of this bacterium via semen.
