A portable surface plasmon resonance (SPR) sensor with a reference channel has been applied to an immunosensor to detect a biomarker, C-reactive protein (CRP) in a human fluid. To detect the biomarker, the sensor chip made of a deposited Au film on a glass chip has been properly modified by sequence reactions of the self-assembled monolayer (SAM) formation, antibody immobilization and non-specific binding block. For the optimal preparation of a bio-recognition layer on the sensor chip, the saturation value of the antibody of CRP (Anti-CRP) concentration has been studied. In addition qualitative and quantitative information on formation of the SAM, immobilization of the antibody, blocking of non-specific binding and interaction of antibody antigen have been investigated by measuring the change in the SPR response during each reaction. The experimental result demonstrates the feasibility of the SPA as a diagnostic tool.
