Alfin-like transcription factor family: characterization and expression profiling against stresses in Brassica oleracea

Brassica oleracea is an important vegetable crop worldwide. The yield and quality of B. oleracea can be threatened by both biotic stresses and abiotic stresses, such as cold, salinity, and drought. Alfin-like (AL) transcription factors (TFs) are important regulators for the process of plant development, but their responsiveness to environmental stresses has not been characterized, except for salt stress in Medicago sativa also called alfalfa. Here, we searched the Bolbase database and identified 12 AL TFs. A phylogenetic tree was constructed based on AL protein sequences of different crops and categorized into four different groups. The corresponding BoAL genes showed organ-specific expression, and genome mapping revealed that BoAL genes were distributed on six out of the nine B. oleracea chromosomes and that 50 % of the family members were segmentally duplicated genes. Furthermore, ten BoAL genes were significantly expressed in response to abiotic stresses, such as cold (4 degrees C), salinity (250 mM NaCl solution), drought (keeping the seedlings on the filter paper at 28 degrees C), and abscisic acid (100-mM ABA) treatment for 24 h, and two BoAL genes showed significant expression in B. oleracea after inoculation with Pectobacterium carotovorum subsp. carotovorum. Overall, two BoAL (BoAL 8 and 12) genes showed responsiveness to all tested abiotic and biotic stresses. The highly expressed BoAL genes identified here could be potentially utilized in the breeding program of Brassica toward the development of stress resistant cultivars. Our results also provide data sets to functionally characterize this prospective gene family.
