A miniaturized immunosensor platform for automatic detection of carcinoembryonic antigen in EBC

A miniaturized platform using Love Wave surface acoustic wave (SAW) immunosensors was developed to improve the automatic and online detection of carcinoembryonic antigen (CEA) in exhaled breath condensate (EBC). For the convenient measurement of sensor response signal, measurement circuit and sensor interface were implemented. The detection approach, based on conventional sandwich immunoassay using antibody labelled with gold nanoparticles (AuNPs) and subsequently mass enhancement using gold staining solutions, was integrated on the SAW sensor surface. This platform showed a good performance to various CEA concentrations, with a limit of detection of 1.25 ng/mL, and the coefficient of determination (r(2)) of 0.998. A comparison between this platform and a commercial instrument used in clinic was also performed, and the correlation coefficient reached 0.999, demonstrating the high accuracy of the proposed platform. Since CEA in EBC is a potential biomarker for early detection of lung cancer, this platform provides a promising tool for a rapid and non-invasive screening to the population at a high lung cancer risk.
