A novel fluorescent dual-labeling method with fluorescein (FR) and rhodamine B (RB) was used for anti-counterfeiting pelleted seeds of tobacco (Nicotiana tabacum L.) cv. MS Yunyan85 (MSYY85) and Honghua Dajinyuan (HHDJY). Seeds were pelleted using a layer of bentonite, which was sprayed with 2.0 mg ml(-1) solution RB, followed by application of FR in a second layer at a rate of 1 g per 10 g of naked seeds mixed with talc. Control seed were pelleted using bentonite sprayed with water before the talc layer was added. These two fluorescent compounds had no negative effects on the germination and seedling growth of pelleted seeds, and even significantly enhanced seedling dry weight, POD activity and contents of chlorophyll and carotenoid in MS YY85 and increased shoot height and chlorophyll content in HHDJY as compared with the control. In addition, the cotyledon tip represented bright green fluorescence under illumination of blue light (495 nm) due to FR, and meanwhile, the cotyledon vein of the same seedling showed obvious red fluorescence under green light (546 nm) based on the RB. Thus, it indicated that the new dual-labeling method that RB and FR simultaneously applied in the same seed pellets will improve anti-counterfeiting technology and enhance seed security.
