By using headspace solid-phase micro-extraction (HS-SPME), together with gas chromatography and mass spectrometry, the volatile compounds of weak-gluten (strong gluten) wheat sample were extracted, identified and analyzed. Using 50 mu m divinylbenzene/carboxen/polydimethylsiloxane (DVB/CAR/PDMS) coating assembly fiber, experimental conditions including extraction time, temperature, sample amount and resolution time, have been optimized. The results showed that the optimized pretreatment conditions were sample quantity, 20 g, extracting for 60 min at 75 degrees C and desorpting for 5 min at 260 degrees C. The main volatile compounds in wheat by this identification method were hydrocarbons, aldehydes, and then alcohols, ketones. The total volatile matter content of wheat decreased firstly and then increased during the 6 months storage. The relative content of hydrocarbons in weak gluten wheat increased rapidly, that of aldehydes decreased firstly and then increased, and that: of ketones and alcohols decreased gradually as storage time prolonging. To the strong gluten wheat, the changing law of the other volatile matter content was similar as the weak gluten wheat except that the relative content of hydrocarbons decreased firstly and then increased rapidly. The wheat volatile matters with more significant changing after 6 months normal temperature storage were 1-hexanol, hexanal 2, 6,10-trimethyl-dodecane, pentadecane and eicosane.
