A novel method for the continuous production of gamma-aminobutyric acid (GABA) using immobilized rice bran was developed. Calcium alginate gel was used to entrap rice bran directly and orthogonal array design was used to obtain the optimal conditions for immobilization. Under these conditions, the preservation of glutamate decarboxylase (GAD) activity was 48.94%. The optimum temperature for immobilized rice bran GAD activity was found to be 45 degrees C with the optimum pH at 5.5. The thermostability, pH stability, operation and storage stability of GAD were improved by immobilization on rice bran. Based on these characteristics, a column reactor of immobilized rice bran was applied to produce GABA continuously. The GABA produced in 60 h by this method was 88.11% of the GABA produced by the free rice bran method, while the protein and sugar contents of the product were greatly reduced. The high output and purity of the product permit its biotechnological potential to be exploited in food applications.
