Molecular cloning and functional characterization of a DNA damage-inducible (DDI) gene in Arabidopsis

DNA damage can affect cell function and survival in living organisms and even transfer to the next generation on the genetic level. DNA damage repair plays important role in maintaining genome integrity. In this study, a DNA damage-inducible gene, designated as AtDDI1, was isolated from Arabidopsis using RT-PCR method. The results of sequences alignment showed that AtDDI1 is well conserved among different plant species. The transcripts of AtDDI1 were expressed constitutively in different tissues, with higher expression occurring in the roots and developing seeds. AtDDI1 expression level was increased after SA, PEG and NaCl treatments in leaves of Arabidopsis seedling. Furthermore, AtDDI1 was significantly induced by the infection of facultative fungal pathogen Fusarium graminearum and the biotrophic fungal pathogen Erwinia carotovora subsp. carotovora. Attenuating AtDDI1 in Arabidopsis through T-DNA insertion increased susceptibility to E. carotovora subsp. carotovora and reduced expression of the SA related genes, while did not affect the expression levels of JA related genes. Also, SA levels in AtDDI1-deleted Arabidopsis decreased significantly. These results suggested that AtDDI1 may play a role mainly in SA-dependent defense response.
