Effects of leptin in rat liver antioxidant systems

Objective: The aim of this study was to investigate the effects of exogenous leptin administration on liver tissue antioxidant system activities in cold-restraint stress and the effect of leptin against the activities of these systems. %26lt;br%26gt;Methods: In this study; 31 Wistar albino male rats (200 +/- 20 g) were divided into 4 groups: 1. control, 2. stress, 3. leptin, 4. leptin+stress. Stress and leptin groups were injected PBS/leptin (i.p., 10 mu g/kg/day) twice for 7 days followed by an additional 4 hours cold-restraint stress at +4 degrees C. After the sodium-thiopenthal administration, animals were immediately sacrificed. The liver glutathione peroxidase (GPx), thioredoxin reductase (TR) and superoxide dismutase (SOD) activities were determined. %26lt;br%26gt;Results: GPx activities in the stress group and the leptin group were significantly reduced (p %26lt; 0.001 and p %26lt; 0.004, respectively) when compared to the control group. GPx activity in the leptin group was significantly increased (p %26lt; 0.021) in comparison with the stress group. A significant GPx activity increase (p %26lt; 0.004) was observed in the leptin+stress group when compared to the leptin group. TR activities in the leptin and leptin+stress groups were significantly reduced (p %26lt; 0.005 and p %26lt; 0.002, respectively) in comparison with the control group. However, leptin group TR activity was not significantly different (p %26gt; 0.05) than the stress group. SOD activity was increased significantly (p %26lt; 0.001) in the leptin+stress group when compared to the stress group, whereas there was no significant difference (p %26gt; 0.05) among the other groups. %26lt;br%26gt;Conclusion: In this study, it was observed that stress damages oxidative balance in liver. Leptin may protect liver antioxidant enzymes.
