The aim of this investigation is to design a novel small intestine-specific delivery system of antidiabetes mellitus by coating chitosan on SBA-15 functionalized with COOH groups, and metformin hydrochloride is used as a model drug to study the drug release kinetics of the carriers. The drug loading capacity of the SBA-15-COOH (52.3 wt%) is more than twice as much as the SBA-15 (25.3 wt%). The excellent loading capacity for metformin hydrochloride is attributed to the specific interactions between the -COOH of SBA-15-COOH and the -NH2 of metformin hydrochloride. The release in vitro shows that the metformin hydrochloride is released from Chit/SBA-15-COOH is retarded at low pH 4.0, which is low drug (18 wt%) released. While at pH 7.4, the release rate of metformin hydrochloride could reach 70 wt%, suggesting that the Chit/SBA-15-COOH will be promising for site-specific drug delivery in the intestine. @@@ A spherical SBA-15-COOH was prepared by SBA-15 functionalized with COOH groups, and the drug loading capacity of the SBA-15-COOH can reach to 52.3 wt%. After coating the Chit, the system shows a pH sensing control release ability. At low pH condition, there is low drug release; while at high pH condition, the Chit/SBA-15-COOH shows the excellent release profile, suggesting that the Chit/SBA-15-COOH will be promising for site-specific drug delivery in the intestine.
