The increasing threats of multidrug resistant fungal pathogens, several studies have been focused to identify novel antifungal plant peptides with unique characteristics. Plants have been defending themselves against fungal infection by generating effective antifungal molecules. Here, a novel antifungal peptide with molecular mass of 4.0 kDa was purified from the pericarp of D.stramonium using reversed phase chromatography system after acetic acid extraction. Presence of sugar moieties (-GlcNAc-) in the peptide have been confirmed using thin layer chromatographic (TLC), CD polarimeter, and MALDI MS analysis. Complete amino acid sequences of this peptide by PSD MALDI MS analysis revealed to contain hydroxyproline in the centre of two cysteine residues. After sequencing TFPKCAPTRhyPhy PGPKhyPCDINNFKSKFWHIWRA-(GlcNAc-)Asn peptide named as datucin. Antifungal sensitivity of datucin have been performed for both planktonic cells and biofilm phenotype of a multidrug resistant clinical isolates, C. albicans and showed a MIC and MBEC values of 1 mu M and 2 mu M, respectively. Hence, D. stramonium offers a potential source of novel antifungal peptide datucin with possible utility in antifungal chemotherapy.
