A new method for the analysis of cyclic peptides in Pseudostellaria heterophylla ( Miq.) Pax was established by high performance liquid chromatography/electrospray ionization time -of-flight -mass spectrometry (HPLC/ESI-TOFMS). The real samples were separated by a reversed-phase C-18 column using a binary eluent under gradient conditions. Six cyclic peptides were isolated and detected with a diode array detector. The cyclic peptides components were identified with HPLC/ESI-TOF-MS technique. ESI-TOF-MS could offer a higher degree of specificity due to its high resolution, which permits accurate relative molecular mass and fragment ion measurement without lose of sensitivity. The [ M + H] + ions of cyclic peptides in the positive mode were observed for molecule mass information. This method developed has the advantages of simple operation, rapid measurement and accurate determination.
