D-myo-Inositol-3-phosphate synthase (MIPS) catalyzes the reaction from D-glucose 6-phosphate (G-6-P) to D-myo-inositol3-phosphate (MIP), which is the first and rate-limited step of myo-inositol biosynthesis. In this study, mungbean (Vigna radiata (L.) Wilczek var. KPS1) MIPS cDNA (VrMIPS1) was isolated from developing mungbean seeds during embryogenesis. The cDNA of VrMIPS1 comprised 1,779 bp, which encodes 510 amino acids with a predicted molecular weight of 56.5 kDa. The VrMIPS1 protein was highly homologous with those from other plant species, ranging from 87% to 96% homology at amino acid levels. RNA blot analysis indicated that the VrMIPS1 transcripts strongly existed in developing seeds and other mungbean tissues, including stems and roots, but showed lower levels expression in young leaves. VrMIPS1 transcripts were observed in the seeds during the early embryogenesis stages. The maximal transcript levels of this gene were observed from 7 to 9 days after flowering (DAF), after which time it gradually decreased. DNA blot analysis indicated that at least three copies of VrMIPS1 are present in the mungbean genome.
