Selective binding of DNA onto SiO2 under high salt concentration has been developed as a general protocol for DNA purifications. It is essential to correctly measure DNA concentration at high salt solution for evaluate the adsorption ability of solid surface. The standard curve of the concentration of DNA solution containing high concentration of salt was tested, correction coefficient was obtained and a new simple reference method to measure the adsorption of salmon sperm DNA on SiO2 was proposed. In comparison with traditional method, this new method is more direct and appropriated for adsorption isotherm characterization and may be used as the basic methodology for evaluating DNA adsorption carriers.
