Various studies have identified that beta-amyloid (1-42) protein in cerebrospinal fluid is one of the most promising potential biomarkers of Alzheimer Disease. It is of particular interest to establish the diagnosis before reaching the stage of clinical severity. In this study, a nano-protein array based on scanning tunneling microscopy (STM) was developed for the detection of beta-amyloid (1-42). Highly dense Au nano dot array (ca. 60 nm) was fabricated on ITO using anodized nanoporous alumina mask. Fragmented monoclonal antibody against beta-amyloid (1-42) was allowed to immobilize on the Au dots followed by its target protein beta-amyloid (1-42). Au nanoparticle-antibody complex was prepared and then applied to preimmobilized protein array to get the pulse like current peak under STM. The current profiles were derived from the STM with different concentrations of antigen. The proposed technique can be a promising method to construct the highly sensitive immunosensor or a high efficiency protein chip.
