Membranotropic Properties of Conjugates of Block Copolymers of Ethylene and Propylene Oxides with Succinic Acid

The conjugates of block copolymers of ethylene and propylene oxides (Pluronics L61 and L121) with succinic acid were synthesized and characterized. We carried out a comparative study of interactions of the polymers with membranes of HeLa and A549 cells. Adsorption of Pluronic L121 on cellular surface is accompanied by a decrease of the negative zeta potential of cells, whereas conjugates of Pluronics L61 and L121, on the contrary, heighten the cell zeta potential. Incubation of cells with Pluronics in buffer solution leads to an increase of plasma membrane permeability to propidium iodide and ions. The efficiency of the polymers grows in the series: L61, conjugates of L121/L61, L121. Upon culturing, Pluronic L61 induces a decrease of mitochondrial membrane potential that is not observed for Pluronic L121. Conjugates of Pluronics exert weaker effects on the mitochondrial membrane potential and have lower cytotoxicity than Pluronics, presumably due to restraints of the intracellular transport of the modified polymers. The results are of interest for the development of efficient and safe drug delivery systems on the basis of Pluronics.
