Mushroom tissue-based flow-injection fluorescence system for the determination of isoprenaline

A novel, simple and reagentless mushroom tissue-based fluorescence system for the determination of isoprenaline over the range of 3 x 10(-8) to 1 x 10(-5) g ml(-1) combined with flow-injection analysis is described in this paper. Mushroom tissue exhibits the rich activity of polyphenol oxidase (PPO), which was employed as the molecular recognition element. Isoprenaline was oxidized under the catalysis of PPO in a tissue column to produce isoproteroquinone. A trihydroxyindole derivative was then generated through a recomposition process in strongly alkaline medium, which was monitored at 417/520 nm( lambda(ex)/lambda(em)). The detection limit is 1.0 x 10(-8) g ml(-1) (3sigma) with a relative standard deviation of less than 5% (n = 11). The reactor was stable for about 14 days and a complete analysis, including sampling and washing, could be performed in 1.5 min. The proposed method has been applied to determination of the studied substance in pharmaceutical preparations with satisfactory results.
