Nano Regulation of Cisplatin Chemotherapeutic Behaviors by Biomineralization Controls

Following its introduction into clinical treatment of cancer in the 1970s, cisplatin [cis-dichlorodiammineplatinum(II)] has become one of the most widely prescribed anti-tumor agents for many malignancies. However, its chemotherapeutic applications are generally limited by severe side effects such as acute nephrotoxicity and chronic neurotoxicity. A design for enhancing therapeutic efficacy and reducing side effects of platinum-based anticancer agents has attracted considerable attentions. Recent achievements in nanotechnology suggest that nanoparticle-based platinum drug can have unique advantages over small molecular cisplatin to improve drug's passive tumor target. After the nano modification, the drug potentially exhibits prolonged blood circulation time with reduced nonspecific accumulation in normal tissues and increased preferential tumor accumulation due to enhanced permeability and retention (EPR) effect, characterized by microvascular hyperpermeability to circulating nanoparticle and impaired lymphatic drainage in tumor tissues. Therefore, it is expected that an increasing of tumor selectivity of Pt-based drug and a reducing of its side effects would be achieved through an appropriate nano modification.
