Electrochemiluminescence (ECL) is chemiluminescence (CL) triggered by electrochemical technique. It has become an important analytical tool because of its high sensitivity, wide linear range, simple instrumentation setup, short analytical time, good selectivity, stable reagents, and wide analytical adaptability. Since Kenten first applied ECL in nucleic acid detection, many new schemes have been developed. This article presents the recent progress of ECL and its applications in nucleic acid detection from 2003 to May 2012. After a brief introduction to the basic principle and merits of ECL, this review presents the classification of ECL assay for nucleic acid detection, labeling oligonucleotides, the combination of magnetic beads-based ECL with nucleic acid amplification technique and nanotechnology, and the applications of direct immobilized ECL assay and capillary electrophoresis (CE)-ECL assay in nucleic acid detection. The development direction of ECL assay for nucleic acid detection is also prospected.
