A simple and specific method for detection of hepatoma specific GGT by affinity chromatography

Background: Various clinical and laboratory experiments have demonstrated that hepatoma specific gamma-glutamyltransferase isoenzyme (HS GGT) is a specific and sensitive biomarker in monitoring the liver carcinogenesis. Various methods for detection of HS-GGT, such as stage gel electrophoresis or chromatography, have been set up however, these methods are complicated. Methods: To set up a method convenient for detection of HS-GGT isoenzyeme, we established a method of datura stramonium agglutinin (DSA) sepharose affinity chromatography to detect HS-GGT in serum and further investigate its diagnostic value in 154 patients with primary hepatocellular carcinoma and benign liver disease and 21 healthy subjects. Results: Results showed this method could distinguish hepatocellular carcinoma from extrahepatic tumors and benign liver diseases such as liver cirrhosis and hepatitis B. Conclusion: Using DSA sepharose affinity chromatography is a simple method with similar diagnostic sensitivity and specificity compared with other methods for detection of HS-GGT.
