A high performance liquid chromatography. charged aerosol detection (HPLC-CAD) method was established for the simultaneous determination of five bile acids in pulvis fellis suis, pulvis billis bovis, pulvis fellis caprinus and pulvis fellis galli. Components were separated and analyzed on a Waters HSS T3 column (100 mm x 2.1 mm, 1.8 mu m). Acetic acid-acetonitrile-water (0.1 : 30 : 70, V/V) and acetonitrile were selected as the mobile phase A and B with a flow rate of 0.3 mL/min. CAD was equipped as the detector with the atomization temperature of 35.0 degrees C and nitrogen gas pressure of 2.413 x 10(5) Pa. Under the selected chromatographic conditione, the limits of detection and quantification of each bile acid were 6 ng and 30 ng, respectively, while the linear range was 0.6-300 mg/L. The spiked recoveries (n = 6) were 85.0%-97.1%. The method was 3.0 times more sensitive than HPLC-Evaporative Light Scattering Detector (HPLC. ELSD) and had a much wider linear range.
