A General Immunoassay for Sensitive Sensing of Universal Biomarker: Human Tissue Polypeptide Antigen

Human tissue polypeptide antigen (HTPA) is a crucial reference index in the benign events of various cancers. In the present work, a general electrochemical immunosensor was developed based on the advanced nanomaterial-technology for the sensitive determination of HTPA. Three-dimensional ordered macroporous gold films (3D-MGM) were employed as the matrix for the capture of antibodies (Ab(1)) having excellent conductivity. Bifunctional, raspberry-like nanoraspberries (BiR-NRs) were employed to label the signal antibody (Ab(2)) to obtain electrochemical signals of catalysis of hydrogen peroxide (H2O2) which exhibited a higher electrocatalytic activity toward H2O2 and a large surface area for the binding of Ab(2). In this strategy, the signal amplification can be achieved successfully. The designed electrochemical immunosensor based on 3D-MGM and BiR-NRs present excellent applicability in determination of HTPA, with a linear range from 0.001 to 15.0 ng/mL and limit of detection of 0.7 pg/mL. Moreover, the immunosensor exhibits outstanding selectivity, good reproducibility and acceptable stability, which will play a crucial role in basic medical research as well as in clinical diagnostics.
