Background: Giardia duodenalis is an intestinal parasite that causes diarrhoea and malabsorption in children. The parasite also infects AIDS patients with weak immune system.
Objective: The objective of the study was to determine the polymorphisms of giardin gene of Malaysian isolates of G. duodenalis by DNA sequencing technique.
Materials and methods: The study was carried out on six Malaysian isolates of G. duodenalis (11.0, 7304, 6304, M007, 2002 and 6307) from faeces of Orang Asli (aborigine) patients admitted to Gombak Hospital. All the isolates were cultured axenically in TYI-S-33 medium. Primer pair LP2 and RP2 were used to amplify the giardin gene by polymerase chain reaction. This primer could detect G. duodenalis to the species levels specifically. PCR products were then sequenced.
Results: A phylogeny tree was drawn using software from clustal-W (Pearson format) from EMBL-EBL. The phylogenetic analysis shows 2 clusters.
Conclusion: We conclude that strain 2002 and 110 were similar to the pathogenic strain WB.
