Allergic diseases afflict many people worldwide. Allergic inflammation mediated by mast cells is involved in allergic disorders. To find efficient therapeutic approaches, the anti-allergic inflammatory effects of eupatilin were investigated using RBL-2H3 cells stimulated by DNP-HSA. As a result, eupatilin can reduce histamine release to attenuate acute allergy, and inhibit the secretion of pro-inflammatory cytokines including TNF-alpha, IL-1 beta and IL-4. Further investigations toward relevant mechanisms have revealed eupatilin can ameliorate intracellular calcium overload to attenuate the histamine release. Moreover, the activation in NF-kappa B pathway is blocked by eupatilin to affect the transcription of pro-inflammatory cytokines. Meanwhile, eupatilin can inhibit Caspase-1 to suppress the secretion of some pro-inflammatory cytokines. These results present evidences for eupatilin to ameliorate mast cells-mediated allergic inflammation.
