The polycomb group transcriptional repressor Bmi-1 regulates both normal and cancer stem cells in multiple tissues. In this issue of Cell Stem Cell, Lukacs et al. (2010) report that Bmi-1 also regulates the self-renewal of at least one prostate stem cell population and is involved in prostate cancer initiation and progression.
