A Model of Cerebral Vasospasms and Metabolomic Mapping

Cerebral vasospasm (CV) is a one of the main factors leading to high morbidity and mortality in patients with a severe subarachnoid hemorrhage. Biomarkers of early diagnosis of the vasospasm development and progression are still poorly understood. Use of microdialysis (MD) techniques combined with modern practices in metabolomic analysis can significantly contribute to the understanding of these phenomena. The aim of this study was to design an animal model of the CV for understanding biochemical correlates of CVs, their development and progression. Intrahippocampal infusion of endothelin- 1, a potent vasoconstrictor, in adult rats was used to mimic CV- condition. A range of biomarkers relevant to clinical practice was monitored using the MD and the HPLC- MS analysis. As a result, we developed a suitable model and a selective method of analysis to monitor neurochemical correlates that are highly relevant for future experimental and clinical research.
