The questions regarding to the increase in zinc and copper contents of kidney and metallothionein in response to silver injection were examined. Each rat was intraperitoneally injected once with 0.9% NaCl or silver (10,20 or 40 mg silver/kg b.w.). After silver injection, 63.7 - 73.2 % of the renal silver contents, 63.2 - 67.8 % of the renal zinc contents and 63.9 - 67.4 % of the renal copper contents were found in the cytosol. Although the silver contents in the cytosol and kidneys of silver-injected rats were higher than those of control rats, they did not increase in response to silver injection. The zinc and copper contents in the cytosol and kidneys did not increase following their injection of silver. The distribution profiles of the renal cytosol of silver-injected rats on a Sephadex G-75 column showed that the amount of the increased silver was attributable to the metallothionein (MT) fractions. The silver concentrations in the MT fractions of silver-injected rats were higher than those of the control rats, but they did not increase in response to silver injection. A dose-dependent increase in the zinc and copper concentrations of the MT fractions was not observed in kidneys with increasing doses of silver. These results indicated that zinc and copper contents in kidneys and zinc and copper concentrations in MT did not increase in response to silver injection.
