摘要
Background: Microalbuminuria is an important prognostic marker of diabetic nephropathy. Several methods have been used for determining urinary albumin; however, none of these methods were found to be a high-throughput analysis. Methods: Urine samples (n=90) were tested for albumin content using a method based on AB 580 with 96-well microplate format and the results were compared with the immunoturbidity method. Results: The assay range was 10-200 mg/L (LOD, 5 mg/L; S/N=3). Good precision was obtained, in which the within- and between-day CVs were 2.48% and 9.16%, respectively. Interference tests were satisfactory with recoveries of 95.7% to 105.5%. This method was highly correlated to the method based on turbidimetric immunoassay (r(2)=0.9269). Conclusions: This study suggested that the AB 580 microplate fluorescent assay might be very promising as a high-throughput and low-cost method for determining microalbuminuria.
