In this issue of Molecular Cell, Ye et al. (2012) show an unexpected cytoplasmic assembly of Arabidopsis ARGONAUTE4/siRNA complexes and that siRNA binding could facilitate the nuclear import of mature AG04/siRNA complexes, revealing a key regulatory process during RNA-directed DNA methylation (RdDM).
