Parentage determination in black tiger shrimp (Penaeus monodon) based on microsatellite DNA markers

Parentage determination was a kind of important technology of selective breeding for aquaculture species. In this study, fourteen microsatellite markers were isolated and chosen to estimate the parentage of 100 progeny in ten black tiger shrimp (Penaeus monodon) full-sib families. Simulation analysis indicated that the power of five loci to exclude false parents was 98.9 % and that of six loci over 99.9 % based on allele frequency data. Moreover, the cumulative assignment success rate was 99 % when no parent information was available using six most informative microsatellite markers. Furthermore, the marker panel tested in mixed families, 89.0 % of progeny was correctly assigned to their parental pairs. Based on the microsatellite analysis, it could be quickly and effectively identified different genealogy and sources of P. monodon; moreover, it provided for selective breeding and multiplication release in this species.
