Aspergillus species are ubiquitous fungi that can cause varieties of lung diseases. Early detection is critical for favorable outcomes. The feasibility of using a technetium-99m (Tc-99m)-labeled anti-Aspergillus monoclonal antibody (WF-AF-1) targeting the Aspergillus cell wall for the early detection of Aspergillus fumigatus in the lung was evaluated. In vitro binding study showed Tc-99m-WF-AF-1 preferentially bound to A. fumigatus. Biodistribution and scintigraphy showed significant accumulation of radioactivity in A. fumigatus conidia-instilled lungs. This study demonstrated that Tc-99m-WF-AF-1 directly targeting A. fumigatus may be valuable for early, noninvasive detection, and provides a referable strategy for the detection of other microorganisms.
