Nanoparticle Formation of Ovalbumin with Cationic Oligopeptides

Here, we report the preparation of ovalbumin (OVA) nanoparticles using cell-penetrating cationic TAT peptides. Nanoparticles with a diameter of 50-200 nm were prepared by heat treatment of OVA/TAT mixtures. Their size can be controlled by adjusting the ratio of OVA to TAT and the feed OVA concentration. DSC and CD measurements of OVA/TAT mixture solution suggest that the TAT peptides had no influence on the heat denaturation temperature and the secondary structure of OVA. On the other hand, fluorescence anisotropy measurements indicate an interaction between OVA and TAT in solution and the equilibrium dissociation constants (K-d) for the OVA-TAT was estimated to be 2.7 mu M. In addition, when the mixtures of denatured OVA with TAT were heated, irregular aggregates were obtained. These results suggest that heat treatment of the ion complex between native OVA and TAT is the key for nanoparticle formation. Furthermore, the OVA/TAT nanoparticles were effectively internalized by RAW264 cells. Thus, nanoparticle formation by heat treatment of OVA in the presence of cationic oligopeptides may provide a new strategy for fabricating protein-based drug delivery carriers.
