A common polymorphism in the coding part of the CD19 gene is not associated with atopy

Recently linkage and association of the chromosomal region 16p12-11 with enhanced IgE responsiveness have been shown. The gene coding for CD19 has been localized to this region. CD19 represents a transmembrane glycoprotein already expressed in early stages of B-cell development; it modulates signal transduction through surface receptors and therefore represents a candidate gene for atopy, We present two common polymorphisms in the coding part of the CD19 gene, one of them leading to an amino acid exchange from arginine to histidine at amino acid position 496 of the gene product, A group of 256 largely atopic individuals has been genotyped for the polymorphism A496H. Association studies revealed no obvious influence on IgE regulation, We conclude that CD19 polymorphisms are unlikely to count for the observed linkage effect at 16p12-11. Whether this polymorphism A496H adjacent to an active binding site of the protein alters the normal functioning of CD19 and influences other immunologic pathways remains to be elucidated.
