In vitro site-specific mutagenesis was used to investigate the mutagenic properties of oligonucleotides containing pyrophosphate internucleotide bonds. The presence of this type of bond in an oligonucleotide did not induce any mutations, but it did promote the fixation of marker deletions specified by the nucleotide sequence. Homozygotes predominated among the mutant offspring, showing that mutation fixation was largely due to a repair mechanism.
