A new, rapid spectrophotometric quantitative determination method for gamma-decalactone (GDL) was developed based on the ferric hydroxamate reaction. Samples added to a mixture of NH2OH center dot HCl and NaOH were heated to react. Then HCl was added. Ferric chloride was added for coloration, and 75% (v/v) ethanol was added. Absorbance values were measured after coloring for 10 min. The concentration range of a GDL calibration curve was 0.125-8.14 g/L. A 96-well plate high-throughput screening method was established to obtain desirable strains. A total of 215 GDL-producing strains were identified from 4,327 samples. The highest production was 179 mg/L.
