Tunisian Nigella sativa seeds were studied for their biological activities. Essential oil was analysed by gas chromatography and gas chromatography/mass spectrometry, whereas the polyphenolic composition was determined by reverse phase high performance liquid chromatography. p-cymene (56.8%) was the major compound in the essential oil, while vanillic acid was the predominant phenolic compound (190 +/- 0.34 mg/100 g dry weight) in the methanol extract. Antioxidant activity was evaluated using 1, 1-diphenyl-2-picrylhydrazyl (DPPH) and superoxide anion radical scavenging. Methanol extract and essential oil were both strong radical scavengers with the essential oil having the highest activity. Antimutagenic activity was determined by the Ames test. Essential oil and methanol extract showed similar important antimutagenic effects; the inhibition percentages were 58.5 and 60%, respectively.
