Molecular analysis of a fragment of the gC glycoprotein of BHV-1 virus isolated from calves in eastern Poland

The aim of this study was to perform the molecular analysis of the glycoprotein gC of BHV-1 virus isolated from calves in eastern Poland. The analyzed material consisted of 191 nasal swabs taken from calves with clinical symptoms of bronchopneumonia. DNA of 1; BHV-1 was detected by the SYBR Green Real-Time HRM PCR technique in 45 swab. Fifteen randomly selected PCR products of Real-Time PCR were sequenced, and the sequences obtained were compared with each other and with the sequence of the gC gene of the It BHV-1 strain from the NCBI GenBank online database (accession number Z49223). On the basis of the analysis of the sequences obtained, three polymorphic variants of BHV1 (No. 1-3) with homology of 99.7-100% were distinguished. The nucleotide and amino acid sequences of the most frequently represented polymorphic variant 1 differed from the nucleotide sequence of polymorphic variant 2 by two substitutions, and from variant 3 by one substitution. The amino acid sequence of the gC gene of BHV-1 (Z49223) differed from analogous sequences of the representatives of variant 1 by 11 substitutions and 3 insertions. Slight nucleotide differences in the gC BHV-1 gene among the viruses suggest that infections in calves were caused by a relatively genetically stable virus that occurs in eastern Poland.
