A reversed phase high performance liquid chromatogaphic method for the determination of organic acid in succinic. acid fermentation broth of Actinobacillus succinogenes was developed. Eclipse XDB-C-8, column (4.6 nun i.d. x 150 mm,5 mum) was used and mobile phase was 0.005 mol/L sulfic acid solution (pH 2.5) with flow rate of 1.0 mL/min. The standard solution of six organic acids (formic acid, malic acid, lactic acid, acetic acid, fumaric acid, succinic acid), was completely separated and determined with variable wave detector at 210 nm within 7 min. The samples of succinic acid fermentation broth would be determined after they were pre-treated by centrifuging. The regression equation, the linear range, the correlation coefficient and the detection limit of the standard curves were determined. The relative standard deviation ( RSD, n = 5 was in the range of 0.5% - 0.9%, and the average recovery was in the range of 99.9% - 101.0%. This method was proved to be a rapid, effective way to determine the organic acid in succinic acid fermentation broth of Actinobacillus succinogenes.
