Introduction: This study was undertaken to investigate the lithium induced restoration of neural stem cells (NSCs) in an in vitro hypoxia model. Methods: NSCs were isolated from neonatal SD rats; subsequently NSCs of third generation underwent hypoxia for 30 min, 1 h or 2 h, and morphological observation, trypan blue staining and CCK-8 assay were done to determine whether the in vitro hypoxia model was successful. Hypoxia-treated NSCs were divided into two groups and treated with normal saline and lithium chloride solution at different concentrations ( I mm, 3 mm). The morphological observation and CCK-8 assay were performed to investigate the NSC induced restoration. Results: NSCs were maintained in glucose free medium in an environment with 95%N2+5%CO2, which could mimic the NSC hypoxia in vitro. When compared with normal saline, the number of NSCs in lithium chloride group was higher, the refractivity was more evident, and the viability and proliferation of NSCs were also elevated (P<0.05). Conclusion: Lithium chloride may protect NSCs against hypoxia.
