A method of hydrophilic interaction chromatography-tandem mass spectrometry (HILIC-MS/MS) was established for the simultaneous determination of five kinds of biogenic amines (cadaverine, histamine, phenylethylamine, tyramine and tryptamine) in bonito. The five biogenic amines were extracted by matrix solid phase dispersion (MSPD), then the extract was separated on a HILIC column with the mobile phases of acetonitrile and 50 mmol/L ammonium formate (pH 4.0 adjusted with formic acid) in a gradient elution mode. The electrospray source was operated in the positive ion mode, and monitored in the multiple reaction monitoring (MRM) mode. The linearity of five biogenic amines was good with the concentration between 0.005 to 0.200 mg/L, and the limit of quantification of the five biogenic amines was 0.1 mg/kg. The recoveries ranged from 74.5% to 116.5% with the relative standard deviations in the range of 3.8%-13.9% (n=6). The results indicated that the method was simple, rapid and accurate for the determination of five biogenic amines in bonito without the need of derivatization.
