Partial Purification and Characterization of Cellulases Produced by bacillus Strain

摘要

The present study deals with the partial purification and characterization of cellulases. Cellulases produced from wild Bacillus PC-BC6 and its mutant derivative Bacillus N3 was partially purified by using 80% ammonium sulphate. An enzyme purification factor of 2.5 and 2.7 with final specific activity of 110U/mg and 148U/mg protein was obtained from cellulose produced by wild and mutant strain respectively. The partially purified enzyme from both strains showed an optimum temperature of 50 degrees C and broad activity and stability range of pH from 5.0-9.0 and 6.0-9.0 respectively. Among metal ions except MnSO4 and CaCl(2)in case of both wild and mutant strains, respectively all other metal ions show inhibitory effect. SDS and EDTA enhance the activity of enzyme produced from wild and its mutant strain respectively.
