摘要

Parkinson's disease is a common neurodegenerative disease characterised by the loss of dopaminergic neurons in the substantia nigra pars compacta and the formation of a-synuclein aggregates found in Lewy bodies throughout the brain. Several a-synuclein transgenic mouse models have been generated, as well as viral-mediated overexpression of wild-type and mutated a-synuclein to mimic the disease and to delineate the pathogenic pathway of a-synuclein-mediated toxicity and neurodegeneration. In this review, we will recapitulate what we have learned about the function of a-synuclein and a-synuclein-mediated toxicity through studies of transgenic animal models, inducible animal models and viral-based models.
