摘要
In this study, a simple, rapid and sensitive high performance liquid chromatography (HPLC) method is developed for determination of penciclovir in human plasma samples from bioequivalence assay. Sample preparation was accomplished through a simple precipitating protein procedure with 70% perchloric acid, and chromatographic separation was carried out on an Agilent ZORBAX-SB-C18 (5 x 150 mm, 5 mu m) at 35 degrees C. Mobile phase composed of a mixture of acetonitrile-3% glacial acetic acid-water (2.5:10:87.5) at a flow rate of 1.0 mL/min. Wavelength was set at 254 nm. The retention time for penciclovir was approximately 4.626 min. The method herein described was superior to previous methods and was successfully applied to a bioequivalence study of oral of famciclovir in Chinese healthy volunteers.
