Development of a simple pyrene-based ratiometric fluorescent chemosensor for copper ion in living cells

A simple pyrene compound (1) containing thiophene is synthesized and used as ratiometric fluorescent chemosensor to recognize Cu2+. The ratiometric signal change of the chemosensor is on the basis of monomer-excimer conversion of 1 induced by Cu2+. As far as we know, it is the first ratiometric chemosensor based on pyrene compound that is developed for Cu2+. The analytical performance characteristics of the proposed Cu2+-sensitive chemosensor are investigated. The chemosensor can be applied to the quantification of Cu2+ with a linear range covering from 1.0 x 10(-7) to 2.0 x 10(-5) M with the detection limit is 2 x 10(-8) M. The experiment results show that the response behavior of 1 toward Cu2+ is pH independent under a wide range of pH from 3.0 to 8.0. Most importantly, the fluorescent changes of the chemosensor are remarkably specific for Cu2+ in the presence of other metal ions, which meet the selective requirements for practical application. Moreover, the response of the chemosensor toward Cu2+ is fast (response time less than 2 min). In addition, the chemosensor has been used to detect Cu2+ in water samples and image Cu2+ in living cells with encouraging results.
