Black fly larvae remain unknown because they cannot be associated with the known species described from other life stages. In this study, mitochondrial cytochrome c oxidase subunit I (COI) barcoding sequences were used to associate unknown larvae with the known species of black flies of the Simulium multistriatum-group in Thailand. Phylogenetic analyses and distance-based criteria were used to associate unknown specimens with the known species. The unknown larvae were separated into two monophyletic clades with strong support, one with S. chaliowae and another with S. bullatum. The results are consistent with distance-based species identification that assigned unknown larvae to S. chaliowae and S. bullatum; thus the results strongly suggested that the unknown larvae were S. chaliowae and S. bullatum. Morphological descriptions of these larvae and characters for species diagnosis are provided. Our results indicate that DNA barcoding sequences can be used effectively to associate unknown life stage with the known species, and this ability will facilitate further study of all aspects of black fly biology.
