Angiogenic potential of bioactive glasses ionic dissolution products

Angiogenesis is complex multi-step process that involves endothelial cell activation, dissolution of surrounding basement membrane, increased endothelial cell proliferation and migration, tube formation to form a vascular network. Recent attempts to stimulate angiogenesis have focused on the delivery of growth factors and cytokines, such us vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF), gene therapy and cells-based therapy. However the growth factors are expensive and the optimal delivery strategies are unclear. Therefore use others easier and cheaper strategies to induce angiogenesis could provide and alternative approach to the use of expensive growth factor for stimulating neovascularization of wound repair. While the bioactive glasses have been extensively investigated for bone repair and regeneration, there has been relatively little research on the application of bioactive glass to highly vascularized soft tissues repair and particularly, the effect on the neovascularization. However, there is emerging evidence in the literature that the use of bioactive glasses in biomaterial-based tissue engineering strategies may improve the vascularization, it is the major limitation of tissue engineering approaches for the replacement of diseased or damaged tissue. This article, in the first part, reviews the bioactivity characteristics of silicate bioactive glasses. In the second part, discusses the evidences in the literature showing the angiogenic potential of bioactive glasses ionic dissolution products, focusing on 45S5 bioglass (R).
