Dosimetry study on irreversible electrical breakdown of rabbit liver tissue under steep pulsed electric field

摘要

The application of steep pulse to tumor treatment has been proposed as a promising method. The basic idea of steep pulse therapy is to apply steep pulse to tumors and thus induce irreversible electrical breakdown and apoptosis, namely programmed death, of cancer cells. The application of steep pulse can also induce anti-blood vessel, anti-lymphatic vessel and immune effect and therefore will contribute to destroy life condition of tumors. A lot of research and development work concerning steep pulse therapy has been carried out. It has been proved that steep pulse therapy is an effective and efficient method and thus has a very promising clinical prospect. However, dose-response study of steep pulse therapy is very limited so far. This paper deals with the quantitative study on the parameters of pulsed electric fields and the corresponding damaged area. Studies on lethal effects of steep pulsed electric fields of different parameter combinations (peak value of voltage, pulse width and frequency) on irreversible electrical breakdown of rabbit liver tissue (damaged area) have been conducted. Each pulsed electric field applied to rabbit liver has a different pulse parameter combination. Damaged area is measured and thus the relative dose-response relationship is obtained. Results show that the value of steep pulsed electric fields parameters has a great influence on the damaged area. With the increase of parameter value, a rise in area damaged and also a saturation tendency can be observed. Although the study carried in this paper is quite primary, it can also provide useful information for further clinical studies on steep pulse therapy.
